Mary Jane technically made her first appearance in The Amazing Spider-Man #25 (June 1965). We say technically because a plant obscured her face and wasn't finally revealed until The Amazing Spider-Man #42 (November 1966), she did however appear in the Spiderman films featuring Tobey Maguire being his on-off love interest.
